Mechanism of action of acyclovir
HSV thymidine kinase:
Facilitate cellular uptake and initial
phosphorylation
Cellular enzymes:
cellular GMP kinase converts acyclovir
monophosphate to acyclovir - diphosphate
cellular phosphatases converts acyclovir-
diphosphate to acyclovir - triphosphate

Acyclovir triphosphate inhibits viral DNA synthesis
by two mechanisms:
Competition with deoxyGTP for viral DNA
polymerase , resulting in binding to DNA
template as an non-functional complex
Also incorporated into viral DNA where it acts as
a chain terminator (because of lack of 3-hydroxyl
group)
