Synthesis of Boc-Ala-Leu-OMe (1)

Name Equivalent Molecular Mole Weight taken

weight
Boc-L-Ala-OH 1 189.20 0.0158 3g

H-Leu-OMe 1.5 181.5 0.0237 4.316 g

DCC 1.2 206.33 0.01896 3.91 g

DMAP 1.1 122.17 0.01738 2.12 g
Synthesis

• Boc-Ala-OH was dissolved in

dry DCM in an ice water and
nitrogen is purged into the RB
to avoid oxidation .

• The reaction is carried out

under appropriate conditions.
7
• It was then added to the
reaction mixture followed by
immediate addition of DCC
and DMAP.

• The reaction mixture was

allowed to stir at room
temperature for 24 hr.

8
Filtering using crucible
• After completion of the
reaction, the resultant
product was taken in ethyl
acetate.

• N,N'-dicyclohexylurea (DCU)
was filtered off.
9
Workup
• The general procedures for
isolating different types of
product are called “work-up” .

• The organic layer was washed

with HCl, sodium bicarbonate,
and brine solution.

10
Column
Chromatography
• It is an technique used for
separating a mixture of chemical
substances into its individual
components.
• They all have a stationary phase (a
solid, or a liquid supported on a
solid) and a mobile phase (a liquid
or a gas).
11
Vacuum pump

• It removes gas from an area,

leaving a partial vacuum
behind.

• The vacuum pump will also

remove water from that
particular compound placed
inside the conical.

Rotary evaporator
• A rotary evaporator is a device used
for the efficient removal of solvents
from samples by evaporation.
• The rotovap works by increasing the
rate of evaporation of the solvent by
• (1) reducing the pressure to lower
the solvent boiling point.
• (2) rotating the sample to increase
the effective surface area.
• (3) heating the solution.

Conclusion

• In summary, we have successfully synthesized a

sequence defined tripeptide (Boc-Ala-Leu-Ala-OH) (Ala:
alanine; Leu: leucine). All the synthesized intermediates
were characterized by 1H NMR spectroscopy.
• Attachment of chlorambucil with the Boc-Ala-Leu-Ala-
OH was confirmed by NMR spectroscopy. In future we
will try to inspect the anticancer activity of this peptide
conjugated drug. 18
Micropipette

• Micropipettes are used to

measure and deliver accurate
volumes of liquid.

• The advantage of micropipette

is that micropipettes measure a
much smaller volume, starting
at 1 microliter.
