ISOLATION, SCREENING AND CHARACTERIZATION OF PSEUDOMONAS ISOLATES FROM

RHIZOSPHERES OF TAMIL NADU AGAINST RHIZOCTONIA SOLANI

INTRODUCTION

Sheath blight (ShB) disease is caused by Rhizoctonia solani, and causes the yield loss up to 69%. Fluorescent Pseudomonas have drawn
much attention worldwide because of their plant growth promotion ability and biocontrol potential against plant pathogens. Hence it is
worthwhile to investigate the biocontrol and growth promoting potentials of FPs from different Rhizospheres.

MATERIALS AND METHODS

• Collection of rhizosphere soils


• Isolation of fluorescent Pseudomonas (FPs) from
Green fluorescent colonies of Rhizoctonia solani on
rhizosphere soil Pseudomonas sp PDA media
• Screening of FP isolates against plant pathogen Rhizoctonia
solani
• Effect of culture filtrate of FP isolates on sclerotial
germination of R. solani
• Biochemical identification of FP isolates
• Mechanism of biocontrol by FP isolates – Detection of
Siderophore production by chrome azurol S (CAS)
• Mechanism of biocontrol by FP isolates – Hydrogen cyanide Screening of Fluorescent pseudomonas Isolates against
(HCN) production Rhizoctonia solani

CONCLUSION

• The complete work has indicated a major biocontrol role for


Pseudomonas sp. Isolates against Rhizoctonia solani.
• Hence field trials for the effectiveness of Pseudomonas sp.
Isolates is very promising.
• It can be recommended that the production of Pseudomonas
sp. Isolates can be increased to commercial levels and used
for field applications.
