INTRODUCTION 2

 Micro cracks are the main cause to structural failure.

 As a result water and other chemicals enters in to the


structure.

 It cause corrosion of steel reinforcement.

 One way to repair these cracks is by an automatic


mechanism.

 It is done with the help of calcite precipitating bacteria.

 It precipitate calcite mineral and fill the cracks in the


concrete.

OBJECTIVES
 To study the effects of adding bacteria to concrete.

 To study the strength charatristics in concrete due


to bacterial action.
 To observe the change in density due to the addition
of bacteria.

The journal we selected is “ A STUDY ON STRENGTH

MATERIALS REQUIRED:
 Cement- OPC of grade 53.
 Coarse aggregate
 Fine aggregate
 Water
 BASCILLUS SUBTILIS of strain JC3

 Bacillus Subtilis JC3 bacteria of concentration 108
cells/ml was used.

 Bacteria is diluted at concentration of 103, 105, & 107


cells/ml.

 M30 grade concrete is used.

 Bacteria added at different concentrations.

 The bacteria is collected from the microbiology dept. of


horticulture college, Mannuthy.

 These are then mixed together with the concrete


mixture at different percentage .

 The specimens of M30 grade are made with and without


bacteria.

 Compressive strength test, Splitting tensile strength


tests ,flexural strength test are to be done.

 Then the results are compared.

METHODOLOGY
 A specific group of alkali resistant spore forming
bacteria of genus Bacillus are selected.

 The bacteria we use here is Bacillus Subtilis.

 These are added to the concrete mixture.

 Standard cubes ,cylinders and beams are made.

 Compressive strength, tensile strength etc are done.

 Compare the result with conventional concrete.


ABOUT BACILLUS SUBTILIS 8

 It is an aerobic, spore forming bacteria.

 It also lives in anerobic condition.

 It will become active in the presence of oxygen.

 In unfavourable condition, it form spores and in


favourable condition it will break the spore and get
active.

 Due to it's spore forming ability, it can live about 300


years.

WORKING PROCESS

 Bacteria is supplied with calcium-based nutrient.


 When cracks appears in structure, water and air enters
into the structure.
 Bacteria get active when in contact with moisture and
air.
 Then the calcium source in the bacteria forms calcium
hydroxide

 Ca(OH)2 + CO2 CaCO3 + H₂O

 Ca(OH)2 acts as catalyst.

According to IS10262 ,for 1 m3 M30 concrete:

 cement - 450 kg/m3

 water -197kg/m3

 Fine aggregate – 797kg/m3

 Coarse aggregate – 1048kg/m3

 water cement ratio – 0.42

• Culturing of bacteria 15

 The bacteria ‘BACILLUS SUBTILIS JC3’ was collected from


Agricultural center, Mannuthy.

 Nutrient broth were prepared for the growth of bacterias by


the help of biotechnology department of our college.

 Several processes like sterilization and inoculation of bacteria


was done.

 It is then kept in a shaker for the growth according to the


concentration required.

 Concentration was measured with the help of uv visible


spectrophoto meter.

Conclusion

 From the experiments conducted, we infer that bacterial


concrete can effectively increase the strength of concrete by
closing the micropores in concrete and making it denser.

 The 7th and 28th day strength of all specimens of bacterial


concrete are stronger than conventional concrete.

 The optimum concentration of cells are found to be 107


cells/ml from the experimental results.

 Percentage increase in strength is greater in splitting tensile


strength than other two.
