Safety practices
General
The following safety practices are intended to insure the safe operation of
the equipment.
Electrical hazards
1. Removal of some panels exposes potentially dangerous voltages.
Disconnect the instrument from all power sources before removing
protective panels.
2. Replace blown fuses with size and rating stipulated on the fuse
panel or holder, and in the manual where listed.
3. Replace or repair faulty or frayed insulation on power cords.
4. Check actual line voltage to confirm it is the value for which this
instrument is wired. Be sure power cords are plugged into correct
voltage sources.
General Precautions
1. Perform periodic leak checks on supply lines.
2. Do not allow flammable and/or toxic solvents to accumulate. Follow
a regulated, approved waste disposal program. Never dispose of
such products through the municipal sewage system.
4 Instrument description
Instrument description
General description
The compact and robust Smartline Autosampler 3800 is the ideal
choice for handling large numbers of samples. The sample carousel
accommodates 96 standard 1.8 ml vials. The standard sample tray is
composed of four segments which can be conveniently rearranged or
exchanged. A sample carousel for 5 ml vials with 48 positions is
optionally available. Thanks to its support of communication via its serial
interface, the Smartline Autosampler 3800 can be easily integrated into
almost any chromatography system. When used together with HPLC
software ChromGate® or EuroChrom®, the user can benefit from simple
control and time-saving features. Manual programming of the
autosampler is also possible through the straightforward menu structure
of the integrated control panel.
Maximum injection accuracy and reproducibility are ensured by using full
sample loop filling. Large volumes can also be reliably injected by using
the selectable time-based loop-filling by headspace pressurization.
The injection volume can be adjusted from 5 – 5000 µl by exchanging the
sample loop. Sample carryover is minimized by the selectable needle
wash function.
The available factory-installed options Cool, Bio and Plus can also be
combined, allowing you to configure the autosampler precisely to your
requirements. With the Cool option the sample tray can be maintained at
4°C by way of a thermo-electric Peltier device. The Bio option equips the
autosampler with a PEEK injection valve and special injection needle for
biocompatibility. The Plus option enables variable injection volumes in 10
µl increments without having to exchange the sample loop.

Installation
Unpacking
Inspect the AS 3800 for indications of damage. Damage that occurs to
the AS 3800 in transit, indicated by damaged containers, is the
responsibility of the carrier and should be reported to the carrier
immediately. Shipping containers should be inspected by the carrier if a
claim is filed.
For contents of shipping containers see the packing list in container.
Location of AS 3800
The AS 3800 needs approximately 30 cm (12 inches) of bench space and
one power supply (230 VAC or 115 VAC, 50/60 Hz).
Be sure that none of the ventilation holes are blocked. Blockage of the
ventilation holes can cause malfunctioning of the AS 3800 or even
damage the electronics inside.
Do not install the AS 3800 in areas subject to excessive dust, direct
sunlight or shocks and do not place it near any other source of heat, this
will disturb the cooling of the tray when installed.
Connections
Power connections
Before plugging in the power cable, check voltage setting of the AS 3800
at the input socket on the rear-panel. Make sure that the voltage setting is
identical with the voltage of your local power supply; use only a supply
appliance with protective grounding.
If the indicated voltage is not correct select the proper voltage by
removing, inverting, and then re–entering the voltage selector-cartridge.
Check if the right fuses are installed, if not replace them with the right
fuses.

Fluid connections
The instrument has been tested with isopropanol: make sure that the
mobile phase of your HPLC system is miscible with isopropanol. If not,
start up with an intermediate solvent as mobile phase (disconnect
column). Connect the HPLC pump to port 5 and the column to port 6 of
the injection valve.
To get access to the fluidics, it is necessary to open the front cover of the
AS 3800, therefore remove the screw on the left-hand side of the cover.
Sample loop
The AS 3800 is factory-equipped with a 20 µl sample loop. If a different
volume is to be injected, the sample loop must be replaced. Please note
that 3 x the loop volume will be withdrawn from the sample vial for loops
smaller than 100 µl and 2 x the loop volume, to fill the loop quantitatively.
For partial loop fill injections the maximum injection volume for a loop is
50% of the loop volume.
HPLC connections
To ensure reproducible injections the following connections to your HPLC
system should be made:
Connect the HPLC pump to port 5 of the injection valve.
Connect the HPLC column to port 6 of the injection valve.
The instrument has been flushed with isopropanol: make sure that the
mobile phase of your HPLC system is miscible with Isopropanol. If not
start up with an intermediate solvent as mobile phase (disconnect the
HPLC column)
It's essential that the contents of the sample loop are injected in back
flush onto the column, therefore: Do not exchange column and pump
connections at the injection valve.

Preparation of vials
Vial dimensions
The AS 3800 is standard equipped with a tray which can contain 96
standard sample vials. The positions 92 – 96 are in use for wash
solutions. Without needle wash they also can be used for samples.
Positioning of vials
Sample vials should be placed in sequential positions on the tray. Empty
spaces between vials of a sample series are allowed, since the AS 3800
can detect empty positions. After detecting an empty place the AS 3800
will continue with the next position.
Additional samples can be placed during RUN since LAST VIAL value
can be changed during RUN (see Running on page 20).
Sample consumption
Rest volume:
Since the sample needle is always at some (very short) distance above
the bottom of the sample vial, a certain volume of sample always remains
in the vial, out of reach of the sample needle. This volume is called the
rest volume.
For standard 1.5 ml vials, the rest volume is approximately 200 µl. For a
conical vial, the rest volume is only a few µl (see Needle penetration
depth on page 25).
Injection volume:
Due to the reduced solvent velocity near the wall of the tubing (loop), it
takes more than the nominal loop volume to fill the loop reproducibly:
AS 3800 takes for flushed loop injections twice the loop volume from the
sample vial for a sample loop equal to or larger than 100 µl and three
times the loop volume for a sample loop smaller than 100 µl.
In case of partial loop fill injections (AS 3800-plus only) the programmed
sample volume is aspirated into the sample loop, but because of the
same phenomenon only the half loop volume can be injected.
Injection principle
A six-port high pressure switching valve can perform the injections based
on the flushed loop principle. Loop filling can also be done on a volume
basis, controlled by the digital dispenser system or on a time basis by
headspace pressure on the sample (see System settings on page 19) to
shorten filling time for larger loop volumes. The switching patterns of the
valve in "LOAD" and "INJECT" position are shown in Fig. 8. An injection
is performed as follows:

1. With the injection valve in load position, the coaxial needle pair is
inserted into the sample. Air pressure, applied on the sample
through the outer (air-) needle, will force the sample via the inner
(sample-) needle through the sample loop. The duration of the
headspace pressure is programmable; appropriate duration has to
be determined experimentally.
2. Injection valve switches into inject position. Sample loop is now part
of the HPLC mobile phase flow path: sample is transported to the
column. Analysis time starts.

Operation
Operating the AS 3800 is easy; after programming the SYSTEM
SETTINGS, only a few parameters need to be programmed for regular
automatic processing of a series of samples.
All parameter values are saved in memory after power switch OFF,
except tray temperature setting, if tray cool option is installed: tray cooling
is always OFF after power UP.
Principle
Different programming modes
To simplify regular operation, common injection parameters like "analysis
time" and "first/last vial" are programmed in the RUN PARAMETER
mode. Less frequently altered parameters like "auxiliaries YES/NO" are
programmed in the SYSTEM SETTINGS mode. An additional COOLING
CONTROL mode takes care of the sample tray temperature, when the
option is installed.
OTHER MODES:
RUN mode: automatic processing of samples.
REMOTE CONTROL mode: autosampler control via I/O contacts.
SERIAL RS232 mode: autosampler control via serial interface.
READY FIRST
READY status is the "ground state" of the AS 3800 which is initiated
automatically after power UP. Every mode can be entered from READY
status, except PRIORITY mode, which can be entered only when
AS 3800 is in RUN mode. In addition, RUN PARAMETER and COOLING
CONTROL mode can be entered when AS 3800 is in the RUN mode.
Function codes are used to enter a certain mode except for the RUN
PARAMETER mode which can be entered directly via the PROG/END
key and RUN mode (entered by the START command). Every mode uses
the same function code (F, 0) to exit, except for RUN PARAMETER
mode and RUN mode which are left via PROG/END and STOP
respectively.

Programming
Only four modes contain programmable parameters:
- SYSTEM SETTINGS mode to "set" system parameters.
- RUN PARAMETER mode for regular programming of sample
processing.
- COOLING CONTROL mode to program the tray cooling, if
installed.
- SERIAL RS232 mode for autosampler control via serial interface.
The device identifier is also programmable in this mode.

System settings
With AS 3800 in READY status, key in F, 9 to select SYSTEM
SETTINGS mode. Press PROG/END to enable programming of the
following parameters (use STEP key to go to the next parameter):
LOOP VOLUME; Enter volume of the installed loop; 0-999 µl
SAMPLE VOLUME Enter 0 to select STANDARD sample tray.
(STD/LSV); enter 1 if the optional LARGE SAMPLE VOLUME
tray has been installed 48 vials of 5 ml).
For loop filling on a time base, mount the special drain to allow for
pressure controlled sample transport and to prevent hydrostatic
sample flow which may empty your sample during analysis time
(Ref. page 24)!
NEEDLE WASH; Enter 1 to select needle wash.
(YES/NO) Positions 93-96 are now used as containers for
washing solvent. After every sample vial the
needle and connecting tubing is flushed with
solvent from one of the "wash vials" (alternating
between 93 and 96). Enter 0 if no needle wash is
wanted (position 93-96 can now be used for
sample vials).
AUXILIARIES; Enter 1 if time programmable auxiliary output(s)
(YES/NO) are to be used. (ref. page 8) Parameters will
appear in the RUN PARAMETER mode. Enter 0 if
no auxiliaries are to be used.
MARKER PULSE Enter the pulse length of the inject marker;
LENGTH; 0.1 - 2.0 sec.
Run parameters
With AS 3800 in READY status, press PROG/END to select RUN
PARAMETER mode and to enable programming at the same time.
The following parameters can be programmed (use STEP key to go to
next parameter):
FIRST VIAL; enter position number of the first vial of the
sample series in the sample tray.
1-96 without needle wash,
1-92 with needle wash,
1-48 in case of LSV.
LAST VIAL; enter position number of the last vial of the
sample series in the tray. (see FIRST VIAL for
valid numbers)
INJ/VIAL; up to 9 injections can be taken from the same
vial.
Enter number 0-9.
20 Operation
Running
When programming is ready (Ref. page 18), system is ready (see section
Installation on page 7) and AS 3800 is in READY status, press
START/STOP key to start automatic processing of the samples. During
RUN, AS 3800 will show information relevant to the actual process like:
SEARCH VIAL 45, LOOPFILL, WASHING etc. During analysis time the
display will show the following information continuously (example):
The first line shows that the second injection from vial 15 is being
processed; vial 82 is the last vial to be processed. The temperature of the
cooling tray shown at the end of the first line will only be displayed if the
option is present.
The second line shows the time since the injection. The status of the
auxiliaries, the number will be displayed if the auxiliary is ON. (Only
shown when enabled in the settings)
The first character of the second line will be a blinking r if the AS 3800 is
under remote control or a blinking p if a priority sample is programmed.
It is possible to reprogram RUN PARAMETERS during RUN; simply
press PROG/END to enter the RUN PARAMETER mode. Press
PROG/END to exit RUN PARAMETER mode and to continue the RUN
program.
If you do not return to RUN mode, RUN will stop after finishing the current
injection!
Changes in the RUN parameters will be executed immediately after
returning to RUN mode (although an injection will not be interrupted).
COOLING CONTROL can also be reprogrammed during RUN; press F, 7
to enter COOLING CONTROL mode.
Return to RUN (F, 0) to continue RUN!
Hold
HOLD (press HOLD/CONT) can be used to freeze the time base. During
HOLD, the RUN PARAMETER mode, COOLING CONTROL mode and
PRIORITY mode are accessible to reprogram/check parameters. Exit any
of these modes (if entered) and press HOLD/CONT key to continue RUN.
Analysis time of the current injection has then been extended with the
time that HOLD was active.
Priority sample
A sample run can be interrupted to process an emergency sample prior
to the next sample of the programmed samples series: press F, 2 to enter
the priority mode, press PROG/END key and enter the number of the tray
position where the priority vial has been placed. Press PROG/END to end
programming. Return to RUN (F, 0). Priority vial will be processed
immediately after the current vial. A blinking p (first position of lower
display line) indicates that a priority vial is to be/being processed. After
processing the priority vial, RUN will continue where it was interrupted.
Remote control
When AS 3800 is in REMOTE CONTROL mode it can only be controlled
using the "next injection" and "next vial" inputs (see Communication
connectors on page 8). RUN is executed as programmed, but analysis
time is now infinite. Only a "next injection" input signal will initiate the next
injection from the same vial (or from the next vial when the programmed
number of injections per vial have been executed). A "next vial" input
signal will initiate the first injection from the next vial.
With AS 3800 in READY status, REMOTE CONTROL mode is entered
with F, 3; display will show:

Next injection or next vial inputs will no longer have any effect. Exit
REMOTE CONTROL mode (F, 0) to return to READY status.
Serial interface
AS 3800 can be controlled by a PC using the RS232 interface and the
KNAUER HPLC software packages ChromGate® or EuroChrom®. Press
F, 4 to activate the serial mode. From here press PROG/END to enable
programming of the device identifier, programmable from 10 to 19
(Default: 11). Refer to the software user's manual for operating
instructions.
After use
If AS 3800 is not to be used for a while, flush needle and valve with water
and isopropanol (can be placed as last vials of a sample series) and
loosen pump tubing cassette one turn. Never leave AS 3800 out of use
with a buffer or salt solution in the tubings; crystals may block the flow
path and may damage the rotor seal of the injection valve. Power can be
switched OFF since all parameter values are saved in memory (long-life
non-chargeable battery back-up). Only sample tray cooling is changed to
OFF after power UP.
Installation
Install the LSV-option according to the following procedure:
- Replace the standard installed tubing and needles by the tubing and
needles of the LSV option. Follow the procedure described in section
Needle pair replacement on page 25.
- Replace the standard tray by the LSV tray.
- Install the 500 µl-loop to port 1 and 4.
- Check the needle penetration depth and if necessary adjust it, see
page 25.
- Connect the drain to port 3, if the loop will be filled on time base by
headspace pressure. It is also possible to fill the loop with the DDS;
this will take less sample, but essentially more time.
- Change the SYSTEM SETTINGS;

The last SYSTEM SETTINGS parameter change is necessary for
pressure controlled injections.
If the loop is filled with the DDS, be aware that a larger preflush volume
(minimal 50 µl) has to be programmed to avoid cross contamination,
because of the larger internal diameters of the tubing and needle.

Air needle
To replace air needle (Ref. Figure A.1), proceed as follows:
- Remove sample needle as described in A.4.1
- Unscrew air needle and install new air needle.
- Reinstall sample needle as described in A.4.1
Injection valve
AS 3800 is equipped with a Valco or Rheodyne valve with quick connect
mounting. The next section describes how to exchange the valve
assembly and contains the Valco Technical Note 810: “Cheminert®
Module C2 Cleaning and Rotor Replacement”, the Rheodyne “Operating
Instructions for Model 7739 Valve”.
• Disconnect all tubing from the valve. Only the loop can stay in
place.
• Push the pushers A backward to release the valve.
• Pull the valve out of the instrument while turning the valve counter-
clockwise.

• Use a 9/16 hex driver to remove the socket head screws, which
secure the cap on the valve.

• To insure that the sealing surface of the cap is not damaged, rest it
on the outer face. Or, if the tubing is still connected, leave it
suspended by the tubing.
• With your fingers or small tool, gently pry the rotor away from the
driver.
• Examine the rotor sealing surface for scratches. If scratches are
visible to the naked eye, the rotor must be replaced. If no scratches
are visible, clean all the parts thoroughly with an appropriate
solvent, taking care that no surfaces get scratched. (The most
common problem in HPLC is the formation of buffer crystals, which
are usually water-soluble) It is not necessary to dry the rotor.
Reassembly
• Replace the rotor in the driver, making sure that the rotor sealing
surface with its engraved flow passages is facing out. The pattern
is asymmetrical to prevent improper placement.
• Replace the cap. Insert the two socket head screws and tighten
them gently until both are snug. Do not over-tighten them - the
screws simply hold the assembly together and do not affect sealing
force, which is automatically set as the screws close the cap
against the valve body.
• Test the valve by pressurizing the system. If it doesn’t hold
pressure, the valve should be returned to Valco for repair.

Specifications
• Maximum temperature is 100°C.
• The valve is set to hold 345 bar (5000 psi).
• The wetted surfaces are stainless steel and an inert polymer.
The following standard sample loops are available; each loop is supplied
with fittings for direct connection to the valve:
Maintenance
With normal use the valve will give many tens of thousands of cycles
without trouble. The main cause of early failure, which is seen as a leak
in the valve, is abrasive particles in the sample and/or mobile phase,
which can scratch the rotor seal.
Following is the procedure for changing the rotor seal and isolation seal.

• Remove the three stator screws with the 9/16” hex key.
• Remove the stator and stator ring from the valve body.
• Pull the rotor seal off the pins.
• Remove the isolation seal.
• Mount the new isolation seal with the spring side facing away from
the rotor seal.
• Mount the new rotor seal. The three pins on the shaft assembly fit
into the mating holes in the rotor seal only one way. Mount the seal
with the grooves facing the stator.
• Replace the stator ring so that the body locating pin in the stator
ring enters the mating hole in the body.
• Mount the stator on the valve so the stator locating pin in the stator
ring enters the mating hole in the stator.
• Replace the three stator screws. Tighten each an equal amount
until the screws are tight.
Operating suggestions:
Aqueous buffers and salt solutions
To prevent formation of salt crystals in the valve, flush out the flow
passages and needle port with water after using salt solutions.
Use of high pH solutions
The standard rotor seal is Vespel, a DuPont polyimide, which is
exceptionally wear-resistant. It is, however, susceptible to alkaline attack
and deteriorates rapidly when used with solutions of pH 10 or higher. An
alternative material is available for alkaline applications (consult
Rheodyne or local Knauer dealer).
Clogged valve passages
If valve passages are clogged, open them by removing the stator and
cleaning the passages with a small wire (maximum diameter 0.015 in.).
Calibrating sample loops
Sample loop sizes are designated by nominal values which can vary from
the actual value by as much as 20%. This is due to the 0.001 inch
tolerance of the tubing I.D. (see table below). Since standards as well as
unknowns are usually analyzed with the same loop, knowledge of
absolute loop volume is rarely needed. If the actual loop volume must be
known, it is best to calibrate it in place on the valve, so that the flow
passages in the valve (one in the rotor, two in the stator) are also taken
into account.

Eluent: Deionised water (HPLC quality)
The eluent must be degassed in order to avoid
functioning defects caused by air bubbles.
Test solution: 50 ppm uracil dissolved in deionised water (HPLC
quality)

A restriction capillary to build up a minimum pressure of 15 bar at a
1ml/min flow rate
UV detector (measuring cell: 10 mm or smaller path length)
Recorder, integrator, or chromatography software
(data rate: min. 5 Hz).
The pump and the detector must be inspected according to the OQ's

A system schema as illustrated below is set up for all tests.

Set the 1 vial, filled with 1000 µl of sample solution, in the 3800’s sample
tray to position 1. Start the method series that was programmed on the
AS 3800. 20 µl of test solution is injected. This is repeated 8 times. A
chromatogram similar to Fig. 23 should result.

3. Linearity
The linearity is valid for the plus version only. The system settings and
analytical parameters are determined as explained in test 1.
10, 20, 30, 40 und 50 µl of the Uralic test solution are injected three times
each from vial 1. In order to do this 5 different methods with different
injection volumes must be programmed. The 5 methods are to be used in
series 5-9. Make sure that there is sufficient test solution in vial 1
(minimum 1ml).

Sicherheitshinweise
Elektrische Gefahren
1. Die Entfernung von Gehäuseteilen ermöglicht den Kontakt mit
Teilen, die gefährliche Spannung führen! Ziehen Sie deshalb immer
den Netzstecker vor dem Öffnen des Gerätes.
2. Ersetzen Sie durchgebrannte Sicherungen ausschließlich mit
solchen, wie sie auf dem Gerät und im Handbuch spezifiziert sind.
3. Verwenden Sie nie Netzkabel, deren Isolierung beschädigt ist.
4. Vergleichen Sie die aktuelle Netzspannung mit der Einstellung am
Gerät. Stellen Sie sicher, dass das Gerät mit der korrekten
Spannung versorgt wird.

Allgemeine Beschreibung
Der kompakte und robuste Smartline Autosampler 3800 ist die ideale
Wahl, um große Probenzahlen zu verarbeiten. Sein Standard-
Probenkarussell nimmt 96 Gefäße à 1,8 ml auf. Es setzt sich aus vier
Segmenten zusammen, die zur bequemen Bestückung einzeln
ausgetauscht werden können. Optional ist ein Probenkarussell für 5 ml-
Gefäße mit 48 Positionen erhältlich. Dank seiner Kommunikations-
freudigkeit über die serielle Schnittstelle kann der Smartline
Autosampler 3800 in verschiedenste Chromatographiesysteme integriert
werden. In Verbindung mit der HPLC-Software ChromGate® oder
EuroChrom® profitiert der Anwender von einfacher und zeitsparender
Bedienung. Die manuelle Programmierung des Autosamplers ist mit Hilfe
des integrierten Bedienfeldes ebenfalls leicht möglich.
Maximale Injektionsgenauigkeit und Reproduzierbarkeit sind bei voller
Schleifenfüllung gewährleistet. Durch die selektierbare
Überdruckbefüllung werden auch große Volumina mit hoher
Zuverlässigkeit injiziert.
Das Injektionsvolumen kann durch Einsatz entsprechender
Probenschleifen innerhalb eines Bereiches von 5 – 5000 µl gewählt
werden. Eine zuschaltbare Injektionsnadelspülung schützt sicher vor
Verschleppungen.
Die ab Werk erhältlichen Optionen Cool, Bio und Plus sind kombinierbar
für eine Ausstattung, die genau Ihren Bedürfnissen entspricht. Mit der
Cool-Option kann der Probenteller durch Einsatz einer leistungsfähigen
Peltier-Kühlung auf 4°C temperiert werden. Die Bio-Option erlaubt mit
ihrem PEEK-Injektionsventil und spezieller Injektionsnadel
biokompatibles Arbeiten. Mit Hilfe der Plus-Option können variable
Injektionsvolumina ohne Schleifenwechsel in Schritten von 10 µl realisiert
werden.

Installation
Auspacken
Inspizieren Sie den AS 3800 auf Anzeichen von Beschädigungen. Für
Transportschäden des AS 3800, ersichtlich an Schäden am
Transportgebinde, ist der Transporteur verantwortlich. Benachrichtigen
Sie diesen unverzüglich. Wird ein Anspruch von Ihnen angemeldet, wird
der Transportcontainer vom Transporteur inspiziert.
Für den Inhalt der Sendung siehe Packliste des Transportcontainers.
Vor dem Betrieb soll der AS 3800 sich während einer Stunde an die
Umgebungstemperatur anpassen können.
Wir empfehlen, die Transportverpackung aufzubewahren. Sie ist zu
benutzen, falls ein Versenden des AS 3800, z.B. zu Servicezwecken,
erforderlich wird.
Anschlüsse
Netzanschluss
Überprüfen Sie die Einstellung der Eingangsspannung an der
Anschlussbuchse auf der Rückseite des AS 3800. Stellen Sie sicher,
dass diese Einstellung mit Ihrer Netzspannung übereinstimmt.
Verwenden Sie ausschließlich Anschlusskabel mit Schutzerdung.
Falls die Spannung nicht übereinstimmt, ziehen Sie den
Sicherungsträgerblock heraus und fügen ihn um 180° gedreht wieder
hinein.
Überprüfen Sie, dass die richtigen Sicherungen installiert sind und
wechseln Sie diese gegebenenfalls aus.

Flüssigkeitsanschlüsse
Das Gerät wurde mit Isopropanol getestet: Stellen Sie sicher, dass Ihre
mobile Phase hiermit mischbar ist. Anderenfalls starten Sie mit einem
Interimslösungsmittel, dass sowohl mit Isopropanol als auch mit Ihrer
mobilen Phase mischbar ist. Stellen Sie hierbei keine Verbindung zu
Ihrer Säule her. Schließen Sie Ihre Pumpe an Port 5 und die Säule an
Port 6 des Injektionsventils an.
Um Zugang zum Injektionsventil zu erhalten, muss die Fronthaube
hochgeklappt werden. Entfernen Sie deshalb die Sicherungsschraube an
der linken Seite des AS 3800. Bewahren Sie die Schraube für eventuelle
Transportfälle auf.
Probenschleife
Der AS 3800 ist standardmäßig mit einer 20 µl Probenschleife
ausgestattet. Für andere Injektionsvolumina muss die Probenschleife
ausgewechselt werden.
HPLC Anschlüsse
Zur Sicherstellung reproduzierbarer Injektionen müssen folgende
Anschlüsse zum HPLC System hergestellt werden:
Verbinden Sie die HPLC Pumpe mit Port 5 des Injektionsventils.
Verbinden Sie die HPLC Säule mit Port 6 des Injektionsventils.

Großvolumenvials (LSV)
Nadelwaschvials
Wenn Sie das Nadelwaschen programmiert haben (Systemeinstellungen
auf Seite 54), setzen Sie 4 Vials mit Waschlösung (je 1.5 ml, üblicher
Weise die mobile Phase) auf die Positionen 93-96 (setzen Sie alle 4
Vials; weil sie alternierend benutzt werden).
Probenverbrauch
Restvolumen:
Weil die Probennadel immer (sehr dicht) über dem Vialboden steht, wird
auch immer ein kleines Restvolumen im Vial verbleiben.
Für die standard 1.5 ml Vials, beträgt dieses Restvolumen etwa 200 µl.
Für die konischen Vials beträgt es dagegen nur wenige µl (siehe
Nadeleindringtiefe auf Seite 61).
Vorspülvolumen:
Vor der ersten Injektion, wird Probennadel und der Schlauch (NadelÆ
Injektionsventil) mit Probe gespült (zur Fingerpumpen gesteuerten
Schleifenfüllung siehe Injektionsprinzip auf Seite 49).
Abhängig von den Konzentrationsunterschieden zwischen den Proben
und der tolerierten Verschleppung kann das Vorspülvolumen im Bereich
von 30-999 µl programmiert werden.
Injektionsvolumen:
Wegen der geringeren Fließgeschwindigkeit der Probe in unmittelbarer
Nähe der Probenschleifenwandung, wird mehr als das nominale
Schleifenvolumen zur reproduzierbaren Schleifenfüllung benötigt: Der
AS 3800 nimmt für Vollschleifeninjektionen das doppelte
Schleifenvolumen aus dem Probenvial für Probenschleife ≥ 100 µl und
das dreifache Volumen für Probenschleifen ≤100 µl.
Im Falle partieller Schleifenfüllungen (nur AS 3800-plus Version) wird das
programmierte Probenvolumen in die Schleife transportiert. Wegen des
gleichen Phänomens, kann aber nur die Hälfte des Schleifenvolumens
programmiert und injiziert werden.
Injektionsprinzip
Ein 6-Port Hochdruckschaltventil dient der Injektion mit
Vollschleifenfüllung. Die Schleifenfüllung kann entweder auf
Volumenbasis mit der Fingerpumpe (DDS) vorgenommen werden, oder
vermittels eines Kopfdrucks auf die Probe (siehe Systemeinstellungen
auf Seite 54) auf Zeitbasis, um die Füllzeit insbesondere bei größeren
Schleifenvolumina zu verkürzen. Das Schaltbild des Ventils in der
"LOAD" und der "INJECT" Position ist in Abb. 8 veranschaulicht. Eine
Injektion wird wie folgt vorgenommen:

Die Bedienung des AS 3800 ist einfach; nach dem Programmieren der
SYSTEMEINSTELLUNGEN müssen nur noch wenige Parameter
festgelegt werden, um ein automatisches Abarbeiten einer Probenserie
zu ermöglichen.
Alle Parameter werden gespeichert und stehen auch nach dem Aus- und
wieder Einschalten des Gerätes zur Verfügung. Eine Ausnahme ist die
Probentellertemperatur im Falle einer installierten Cool-Option. Diese
Option ist bei jedem Neustart des AS 3800 in OFF.
Funktionsprinzip
Programmiermodi
Um den normalen Betrieb des AS 3800 zu vereinfachen, werden übliche
Injektionsparameter wie die Analysenzeit, das erste und letzte Vial im
RUN PARAMETER Modus programmiert. Seltener veränderte Parameter
wie "auxiliaries YES/NO" werden dagegen im SYSTEMEINSTELLUNGS
Modus programmiert. Ein Zusätzlicher COOLING CONTROL Modus ist
nur bei installierter COOL Option zugänglich.
ANDERE MODI:
RUN Modus: Zur automatischen Probenbehandlung.
REMOTE CONTROL Modus: Steuerung via I/O Kontakte
SERIAL RS232 Modus: Steuerung via serielles Interface.
READY FIRST
Der READY Status ist der "Grundstatus" des AS 3800, der nach dem
Einschalten des Gerätes initiiert wird. Jeder andere Modus kann von
hieraus erreicht werden. Eine Ausnahme bildet der PRIORITY Modus,
der nur erreicht werden kann, wenn sich der AS 3800 im RUN Modus
befindet. Auch der RUN PARAMETER Modus und der COOLING
CONTROL Modus können aktiviert werden, wenn sich der AS 3800 im
RUN Modus befindet.
Funktionscodes werden zum Aktivieren der Programmiermodi genutzt.
Der RUN PARAMETER Modus wird dagegen direkt mit der PROG/END
Taste und der RUN Modus mit der Starttaste aktiviert. Aus den
Verschiedenen Modi gelangen Sie mit einem einheitlichen Funktionscode
(F, 0) wieder in den READY Status. Ausnahmen bilden wieder der RUN
PARAMETER Modus, beendet mit PROG/END und der RUN Modus,
beendet mit der STOP Taste.

Systemeinstellungen
Aktivieren Sie aus dem READY Status des AS 3800 mit dem
Funktionscode F, 9 to den SYSTEM SETTINGS Modus. Drücken Sie
PROG/END zum Beginnen der Programmierung der nachfolgenden
Parameter und verwenden Sie die STEP Taste um zum nächsten
Parameter zu gelangen.
LOOP VOLUME: Geben Sie das Volumen der installierten
Probenschleife ein; 0 - 999 µl
PROBE VOLUME: Geben sie 0 ein für den STANDARD Probenteller
(STD/LSV) oder 1 für den optionalen LARGE PROBE
VOLUME Teller mit 48 Vials von 5 ml.
INJEKTION MODE: (Nur AS 3800-plus)
(FLUSHED/PARTIAL) Geben Sie 0 ein für die Vollschleifeninjektion oder
1 für die partielle Schleifenfüllung
FLUSH: Geben Sie für eine VOLUMEN geregelte Injektion
(VOL/TIME) (VOL) eine 0 ein oder eine 1 für eine ZEIT
geregelte Injektion (TIME). Diese dient besonders

Runparameter
Aktivieren Sie aus dem READY Status des AS 3800 mit PROG/END den
RUN PARAMETER Modus. Sie können jetzt direkt mit der Programmie-
rung der nachfolgenden Parameter beginnen. Verwenden Sie die STEP
Taste um zum nächsten Parameter zu gelangen.
FIRST VIAL: Geben Sie die Positionsnummer des ersten
Vials der Serie auf dem Probenteller ein.
1-96 ohne Nadelwäsche,
1-92 mit Nadelwäsche,
1-48 im Falle des LSV Probentellers.
LAST VIAL: Geben Sie die Positionsnummer des letzten
Vials der Serie auf dem Probenteller ein. Sie
muss innerhalb der für das erste Vial
genannten Grenzen gleich oder größer als
diese sein.
Betrieb 55
Probentellerkühlung (optional)
Probentellertemperatursteuerung wird im COOLING CONTROL Modus
ON/OFF geschaltet.
Geben Sie im READY Status des AS 3800 den Funktionscode F, 7 ein,
um in den COOLING CONTROL Modus zu gelangen. Drücken Sie
PROG/END, um die Steuerung ON oder OFF zu schalten.
Die Probentellerkühlung hat eine Festtemperatur von 4°C.
TRAY COOLING: Geben Sie 1 ein, um die Probentellerkühlung
(ON/OFF) einzuschalten.
Geben Sie 0 ein, um die Probentellerkühlung
auszuschalten
Nach dem Einschalten des AS 3800 ist Probentellerkühlung immer
in der OFF Position!
Injektionsausführung
Wenn die Programmierung abgeschlossen (siehe Seite 53) und der
AS 3800 im READY Status ist (siehe Installation auf Seite 42) drücken
Sie die START/STOP Taste, um den automatischen Injektionsablauf zu
starten. Während des Laufs werden die aktuellen Statusinformationen auf
dem Display angezeigt, beispielsweise: SEARCH VIAL 45, LOOPFILL,
WASHING etc. Die folgende Abbildung zeigt einen Beispielscreen.

In der ersten Zeile wird angezeigt, dass die 2. Injektion aus Vial 15 erfolgt
ist, das letzte Vial die Nummer 82 hat und die Probentellerkühlung aktiv
auf 4°C steht. Die Temperatur wird nur bei installierter Option angezeigt.
In der zweiten Zeile wird die Analysenzeit angezeigt und, falls aktiviert,
der Status der Auxiliaries. Die jeweilige Nummer wird angezeigt, wenn
der Status ON ist.
Das erste Zeichen in der zweiten Zeile kann ein blinkendes r sein
(remote control) oder auch ein blinkendes p wenn eine Prioritätsprobe
bearbeitet wird.
Die RUN PARAMETER können auch während eines Laufes für die
nachfolgenden Injektionen der programmierten Serie geändert werden.
Drücken Sie einfach PROG/END, um in den RUN PARAMETER Modus
zu gelangen. Nehmen Sie die gewünschten Änderungen vor und drücken
erneut PROG/END, um wieder in den RUN Modus zurück zu kehren.
Ohne diese Rückkehr wird die Bearbeitung der Serie mit Ablauf der
Analysenzeit des aktuellen Laufs beendet.
Die COOLING CONTROL kann ebenso während eines Laufs
umprogrammiert werden. Drücken Sie F, 7 um den COOLING CONTROL
Modus zu aktivieren. Mit dem Funktionscode F, 0 kehren Sie in den RUN
Modus zurück.
Auch hier wird ohne diese Rückkehr die Bearbeitung der Serie mit
Ablauf der Analysenzeit des aktuellen Laufs beendet.
Hold
Durch Drücken der HOLD/CONT im RUN Modus stoppen Sie die
Analysenzeit (Anzeige: HOLD). Durch erneutes Drücken der Taste wird
sie fortgeführt. Die Analysendauer wird dadurch um die Zeit verlängert,
die der HOLD Status aktiv war.
Während des aktiven HOLD Status sind der RUN PARAMETER Modus,
der COOLING CONTROL Modus und auch der PRIORITY Modus
zugänglich.
Prioritätsproben
Eine Probenserie kann für die Bearbeitung einer dringlichen
Prioritätsprobe unterbrochen werden. Drücken Sie F, 2 um in den
PRIORITY Modus zu gelangen, drücken Sie PROG/END und geben die
Positionsnummer ein auf die Sie die Prioritätsprobe gesetzt haben.
Drücken Sie PROG/END zur Beendigung der Programmierung und F, 0
zur Rückkehr in den RUN Modus. Die Prioritätsprobe wird nun
unmittelbar als nächste injiziert. Ein blinkendes p in der zweiten

Die Eingangssignale “next injection” oder “next vial” haben jetzt keinen
Effekt mehr. Beenden Sie den REMOTE CONTROL Modus mit dem
Funktionscode F, 0 und kehren damit in den READY Status zurück.
Serielles Interface
Der AS 3800 kann mit den KNAUER HPLC Softwarepaketen
ChromGate® oder EuroChrom®. über das RS232 Interface eines PC
gesteuert werden. Geben Sie den Funktionscode F, 4 ein zur Aktivierung
des SERIAL Modus. Von hier aus Können Sie nach dem Drücken von
PROG/END die Geräte ID programmieren. Sie kann im Bereich von 10
bis 19 liegen, voreingestellt ist 11. Verfahren Sie sonst nach dem Manual
zum verwendeten Softwarepaket.
Spezielle Displaymeldungen
Während des Betriebes können verschiedene Displaymeldungen
angezeigt werden, die Sie auf gegebenenfalls aufgetretene Fehler
aufmerksam machen. Diese Fehlermeldungen sind im Folgenden
aufgelistet, zusammen mit den geeigneten Verfahrensweisen zu ihrer
Behebung.

Außerbetriebnahme
Wenn der AS 3800 einige Zeit nicht benutzt werden soll, spülen Sie die
Nadel und das Ventil mit Wasser und Isopropanol (kann auch als letztes
Vial der Serie eingesetzt werden) und lösen Sie die Befestigungs-
schraube der Fingerpumpenkassette um eine Umdrehung.
Lassen Sie den AS 3800 nie mit Puffer- oder Salzlösungen in den
Leitungen stehen. Kristallabscheidungen können zur Blockierung
des Eluentenweges und zur Beschädigung der Rotordichtung
führen.
Der AS 3800 kann ausgeschaltet werden, da alle Parameter im Batterie
gespeisten Speicher erhalten bleiben.
Installation
Installieren Sie die LSV-Option wie folgt:
- Tauschen Sie die standardmäßig installierten Probennadel und –
leitungen gegen die der LSV-Option aus. Verfahren Sie dabei wie es
im Abschnitt Nadelpaar auf Seite 61 beschrieben ist.

Nadeleindringtiefe
Um das Restvolumen klein zu halten soll die Probenadel kurz oberhalb
des Vialbodens enden. Zur Justierung verfahren Sie wie folgt (siehe auch
Abb. 16):
- Öffnen Sie die Tastaturhaube.
- Setzen Sie ein unverschlossenes Vial in die Position 2 des
Probentellers.
- Programmieren Sie ein Schleifenvolumen von 0 µl. Drücken Sie
START und schalten Sie den Netzschalter aus, sobald die Nadel
im Vial ist.
- Lösen Sie die Einstellschraube um eine Drehung entgegen dem
Uhrzeigersinn.
- Justieren Sie die Eindringtiefe, indem Sie die Verschraubung (4)
als Griff benutzen.
Die Nadel sollte nicht tiefer als 1 mm über dem Boden eines
Standardvials und nicht tiefer als 2 mm über dem Boden eines
konischen Vials stehen.
- Ziehen Sie die Justierschraube im Uhrzeigersinn wieder fest.
- Schalten Sie das Gerät wieder ein.
Sicherungen
BRANDGEFAHR!
VERWENDEN SIE NUR DIE ANGEGEBEN SICHERUNGEN!
Verwenden Sie für 115 VAC ±15%, zwei 5 AT- Sicherungen (slow).
Verwenden Sie für 230 VAC ±15%, zwei 2,5 AT- Sicherungen (slow).
Trennen Sie das Gerät vom Netz, bevor Sie die Sicherungen
herausnehmen!
Nadelpaaraustausch
Probennadel
Zum Wechsel des Nadelpaares verfahren Sie wie folgt:
- Schalten Sie das Gerät aus.
- Öffnen Sie die Tastaturhaube.
- Lösen Sie die Drehgelenkkopplung der Einspritznadel (siehe Abb.
16), die den PTFE Schlauch mit der Nadel verbindet.
- Lösen Sie die Probenadel von dem Fitting mit dem PTFE-
Schlauch, an dem sie üblicherweise befestigt ist.
- Setzen Sie die neue Nadel ein (Achten Sie auf die Luftdichtung um
die Nadel!).
- Zum Austausch des Nadelverbindungsschlauchs lösen Sie dessen
Verbindung am Injektionsventil.
- Schneiden Sie einen 0,25 mm I.D. Schlauch auf eine Länge von
185 mm. Achten Sie auf einen exakt rechtwinkligen Schnitt, um
Totvolumen zu vermeiden.
- Schieben Sie ein Schlauchende auf die Nadel und verbinden das
andere mit dem Port 2 des Injektionsventils.

- Ziehen Sie die Verbindung nicht zu fest an, sonst kann der
Schlauch abgeklemmt werden!
- Überprüfen Sie die Nadeleindringtiefe und regulieren Sie diese
gegebenenfalls nach, siehe Seite 61.
- Schließen Sie das Gerät wieder an das Netz an.
- Die Nadel muss sich innerhalb des Vialsensors befinden.
- Schließen Sie die Tastaturhaube.
Luftnadel
Zum Wechsel der Luftnadel verfahren Sie wie folgt Einspritznadel (siehe
Abb. 16):
- Entfernen Sie die Nadel wie oben beschrieben.
- Schrauben Sie die Luftnadel ab und ersetzen Sie sie durch eine
neue Nadel.
- Reinstallieren Sie das Nadelpaar wie oben beschrieben.
Injektionsventil
Der AS 3800 ist mit einem Valco oder Rheodyne Ventil ausgestattet. Der
folgende Abschnitt beschreibt den Austausch des kompletten Ventils und
enthält die Valco Technical Note 810: “Cheminert® Module C2 Reinigung
und Rotoraustausch”, sowie die Rheodyne “Bedienungsanleitung für das
Ventil Model 7010”.
Valco Injektionsventil
Bauen Sie das Ventil nicht auseinander, wenn eine Fehlfunktion des
AS 3800 nicht eindeutig auf das Ventil zurückzuführen ist.

Demontage
Siehe Abb. 19, Explosionsdarstellung.
• Lösen Sie mit einem 9/16 Inbusschlüssel die Kopfschrauben, die
die Ventilkappe halten.
• Um Beschädigungen der Dichtfläche zu vermeiden, legen Sie die
Kappe nie mit diesen nach unten ab.
• Nehmen Sie den Rotor vorsichtig vom Antrieb ab.
• Untersuchen Sie die Rotordichtung auf Kratzer. Sind solche mit
bloßem Auge zu erkennen, muss der Rotor ausgewechselt
werden. Sind keine Kratzer zu erkennen, reinigen Sie alle Teile
vorsichtig mit einem geeigneten Lösungsmittel. Am häufigsten sind
Probleme in der HPLC auf die Bildung von Pufferkristallen
zurückzuführen. Diese sind gewöhnlich wasserlöslich. Der Rotor
braucht nicht getrocknet zu werden.

Zusammenbau
• Setzen Sie den Rotor mit den eingravierten Fließkanälen nach
außen in den Antrieb ein. Die Abmessungen sind asymmetrisch
gehalten, um die richtige Position sicher zu stellen.
• Setzen Sie die Kappe auf. Fügen Sie die beiden Kopfschrauben
ein und ziehen sie leicht an. Ziehen Sie sie nicht zu fest an, sie
haben nur eine Haltefunktion und beeinflussen die Dichtungskräfte
nicht. Diese stellen sich automatisch ein, wenn die Kappe am
Ventilkörper anliegt.
• Prüfen Sie das Ventil, indem Sie das System unter Druck setzen.
Wenn es den Druck nicht hält, muss es zur Reparatur an Valco
eingeschickt werden.
Rheodyne Injektionsventil
Kopie der Rheodyne Operating Instructions für Modell 7739
Beschreibung
Das Rheodyne Injektionsventil Modell 7739 ist ein 6-Port Rotationsventil
mit austauschbarer externer Probenschleife für die HPLC. Das Ventil ist
licht zu warten und arbeitet über Jahre zuverlässig. Es ist mit 1/16"
Leitungsanschlüssen versehen.

Wartung
Bei normalem Gebrauch sind mehrere zehntausend Schaltungen ohne
Probleme möglich. Hauptgrund für eventuelle frühere Fehler (zu
erkennen an auftretenden Undichtigkeiten) sind abrasive Teilchen in der
Probe oder der mobile Phase, die zu Kratzern auf der Rotordichtung
führen können.
Zum Austausch der Rotor- und der Isolierungsdichtung verfahren Sie wie
folgt, vergleiche Abb. 21:
• Entfernen Sie die drei Statorschrauben mit einem 9/16”
Inbusschlüssel.
• Entfernen Sie den Stator und den Statorring vom Ventilkörper.
• Ziehen Sie die Rotordichtung von den Stiften ab.
• Entfernen Sie die Isolierungsdichtung.
• Setzen Sie die neue Isolierungsdichtung ein. Die Federseite muss
der Rotordichtung abgewandt sein.
• Setzen Sie die neue Rotordichtung. Die drei Stifte gewährleisten
die Richtige Ausrichtung der Rotordichtung. Die Fließkanäle
müssen zum Stator zeigen.
• Setzen Sie den Statorring auf, so dass der Führungsstift des
Ringes in die entsprechende Bohrung passt.

Es werden jeweils 20 µl der Testlösung injiziert. Dies wird 8-mal
wiederholt. Es sollte sich ein Chromatogramm ähnlich wie in Abb. 23

Der Linearitätstest ist ausschließlich für den AS 3800 Plus gültig. Die
Systemeinstellungen und analytischen Parameter sind wie unter Test 2
festzulegen.
Es werden jeweils 3 mal 10, 20, 30, 40 und 50 µl der Testlösung Uracil
aus Vial 1 injiziert. Dazu müssen am Autosampler 5 Methoden mit
unterschiedlichen Injektionsvolumina programmiert werden. Stellen Sie
sicher, dass sich genügend Testlösung in Vial 1 (mindestens 1 ml)
befindet.
