5 Gerätebeschreibung
5.1 Zweckbestimmung

CAUTION: The incubator must be placed in such a way that the power
cable is easily accessible at all times. The power cable is used to safely
disconnect the device from the power supply.
CAUTION: Removable power cables shall not be replaced by those not
designed for the purpose.
NOTICE: Too high a gas pressure can damage the gas connection or
tubing. The prescribed pressure range of 0.5 to maximally 1.5 bar of
the connected gases must always be observed.
NOTICE: Use only aqua bidest without additives or distilled water of
similar quality, like the sterile and pyrogen-free AEROpart®
( REF 16150).
NOTICE: Use of unqualified gas could lead to a negative impact on
growth of the cell culture. The operator must select the proper mixed-
gas and adjust it to the buffer recipe of the culture medium used or to
the desired application.
NOTICE: Unsuitable room temperature (outside of 18 °C to 30 °C),
exposure to draughts or direct sunlight could lead to faulty temperature
regulation of the incubator and a defect in cell growth.
NOTICE: Danger of humidity accumulation/forming of condensing
water in the incubation chamber if humidified humidification stones are
left inside and the incubator is switched off. Therefore, if the device is
to be switched off or has run without gassing for a longer period,
remove the humidification stones from the incubator.
NOTICE: Automatic flushing after lid opening may be hindered if
incubator is placed in a dark room because of the use of an optical
sensor for this function.
To protect valuable cultures or cells we recommend connecting the
remote alarm.
The Labotect gas monitor can be used for monitoring the pressure of
a gas canister and switching to a reserve bottle.
Overfilling of the humidification stone may cause excess formation of
condensation water in the incubator.

4 Scope of delivery
The CO2 Incubator Labo C-Top is designed and produced to provide optimal and constant environmental
conditions. The parameter for temperature is to be adjusted in a way that biological cultures can be
cultivated inside the incubator.
The interior of CO2 Incubator Labo C-Top has a defined atmosphere concerning the parameters of
temperature, gassing and relative humidity. Due to the stability of temperature, gas concentration and
maximum relative air humidity it is suitable for cultivation of biological material in research and medicine.
The Labo C-Top fulfils the fundamental requirements of medical device directive 93/42/EEC. Due to the
design and the CE marking according to the directive, the special conditions for use in medical laboratories,
clinics and hospitals, especially in the area of assisted reproduction techniques (ART) are fulfilled.
Execution of procedures of reproductive medicine and other clinical applications is only permitted for
specially trained and authorised persons.
NOTICE: For clinical applications, the Labo C-Top may only be used
by specialised personnel and with observation of the regionally valid
legal requirements.
5.3 Principle
The Labo C-Top is optimised for reception of cell culture dishes. The incubation space consists of two
independent incubation chambers (also called tray below). Therefore, opening the lid of one tray has no
influence on temperature, humidity and flow in the other tray.
The chamber is designed in a way that optimal heat contact to the bottom of the cell culture dishes is
ensured.
The design of the bottom contour of the trays allows receiving up to 4 x Nunc™ multi-dish 66*66 or 4 x Petri
dish Ø 60 mm or 10 x Petri dish Ø 35 mm.
Simultaneous loading of different culture dishes is possible.
32
Labotect Labo C-Top
33
Labo C-Top Labotect
Temperature Control
Temperature is controlled separately for each incubation chamber for base plate and lid. Base plate and lid
are heated over the full surface.
Even when the lid is opened, the heated elements are maintained at the set temperature (e. g. 37 °C).
Therefore, the selected incubation temperature is immediately available after closing. This temperature
regulation, the close heat contact of the culture dishes and the small volume of the chamber make it
possible that the temperature in the cell culture dishes is maintained very constant.
Per tray, the temperature can be set differently in the range of 2 °C, e. g. on the right 37 °C and on the left
39 °C. In order to be able to prevent exceeding the temperature in the incubation chamber in case of
malfunction, an independent over-temperature regulator is installed in each tray.
Humidification
The incubation space is humidified by means of a water reservoir in the incubation chamber. A micro-
porous humidification stone is filled with maximally 9 ml of suitable water (only aqua bidest without additives
or distilled water of similar quality) that is heated to 37 °C (set temperature) in the incubation chamber. The
inflowing gas flows over the humidification stone, thus distributing the humidity evenly in the incubation
chamber.
Depending on the set gas flow and the number of openings of the incubation space, 9 ml of suitable water
are sufficient to humidify the chamber for up to 4 days.
When the water in the reservoir is running low, an alarm message is generated (rH-L).
This method of humidification has no open water surface. The large porous surface of the humidification
stone humidifies the inflowing gas maximally. This happens independently of the quantity of water currently
available in the humidification stone; i. e. as long as there is water in the humidification stone, the incubation
chamber will be supplied with a maximum of relative air humidity.
A slight film of humidity in the area of the lids of the incubation chambers is normal, due to the high relative
air humidity. This is a physically determined procedure therefore a set-point for relative humidity does not
need to be indicated.
The humidification stone can be sterilised in an autoclave for 20 minutes at 121 °C with fractionated
vacuum, or for 3 minutes at 134 °C with fractionated vacuum.
We recommend carrying out sterilisation of the humidification stone weekly, or after changing the charge
of the incubator. In order to ensure continuous operation, each Labo C-Top incubator is supplied with two
pairs of humidification stones.
34

Gas Supply
To maintain the desired pH value of the medium, the Labo C-Top incubator is gassed with premixed gas
(e. g. 6 % CO2, 5 % O2, 89 % N2).
The gassing rate can be selected by making settings in the operating field between 20 ml/min and
30 ml/min.
After opening the incubation chamber and closing the lid again, the
incubation chamber is flushed for 3 minutes with 250 ml/min. After this
time, the chamber is evenly filled with the premixed gas and the gas
flow is reduced to the defined set-point.
Gas flows from the gas inlet strainer (next to the humidification stone) around the humidification stone
through the chamber and at the outer side out of the incubator.
To regulate the gassing rates, each chamber is equipped with a gas quantity meter and a proportional
valve.
Optional, the Labo C-Top is available with a second gas connection for use of different gases in each
chamber.
Operation
The lid top of the Labo C-Top is designed as glass operating panel. It includes the display and below the
display sensor buttons assigned depending on context for operation of the device (see Figure 1).

Only use a suitable marker pen for labelling on the lid in order to avoid
scratching or irreversible labelling. For the same reason, remove the
labelling only with a soft cloth.
Safety Concept
Emphasis was placed on implementation of a safety concept. The following safety functions are
implemented:
- Monitoring of the sensor signals
- Monitoring of exceeding minimum and maximum set values
- Independent temperature regulators as over-temperature protection
- Monitoring of the lid opening
- Acoustic and visual alarm system
- Storing alarm messages for review
- Remote alarm via potential-free contact
Using the existing RS485 interface, the device can be connected to the measuring value capturing system
LaboDat+. The software LaboDat+ allows representation, backup and printout of data.
36
Labotect Labo C-Top
The LaboDat+ system for capturing measuring values is able to connect up to 16 Labotect units (incubators
and gasmonitor) via RS485-bus to a PC. The software allows representation, backup and printout of the
available operating parameters. The data are protected against manipulation; they are administered with
the highest level of security.
Connections
The following connections are on the rear wall of the device (see Figure 2):
Figure 2: View of the back side of the Labo C-Top incubator with the connections for supply, remote alarm
and the RS485 interface
In order to be able to guarantee the safety and quality of the incubator, the following standards and
directives are observed:
- EN 61010-1:2010 (3rd Edition) und EN 61010-2-010:2014 safety provisions for electric measuring,
control, regulating and laboratory apparatus
- DIN 58945 breeding cases for microbiological purposes
- EC Directive 93/42 regulating medical products

6 Operating Instructions
6.1 Setup
CAUTION: The incubator must be placed in such a way that the power
cable is easily accessible at all times. The power cable is used to
safely disconnect the device from the power supply.
CAUTION: Removable power cables shall not be replaced by those
not designed for the purpose.
NOTICE: Too high gas pressure can damage the gas connection or
tubing. The prescribed pressure range of 0.5 to maximally 1.5 bar of
the connected gases must always be observed. Normal operating
pressure is 0.8 bar.
NOTICE: Setup with clearance to walls and adjoining devices at least
10 cm in order to open the lid fully and to avoid bending supply lines.
NOTICE: Supply pipes must not become buckled and the power
switch on the back of the device must still be accessible.
Preparations
The incubator has passed all tests for quality assurance and to ensure electrical safety. Furthermore it was
tested for operative functionality (cf. supplied test record). Before being packed, the device has been
cleaned and disinfected. The device is supplied ready-to-connect.
Before using the device for the first time, carry out a wiping
disinfection of the exterior and interior surfaces of the incubator in
order to remove dirt from transport and setup. We recommend the
use of Fermacidal-spray.
Before using the device for the first time, autoclave the humidification
stones.
38
Labotect Labo C-Top
In order to set the gas connection pressure of 0.8 bar exactly, only use a pressure reducer with an output
pressure range of maximally 2.5 bar.
For almost full removal of organic contaminants (Oil vapours, odorous substances, aromatic compounds
aldehydes, ketones, pyridines, furans, CHC and others) from incoming gases we recommend the use of
the included Gas In-Line-Filter(s).
Proceed as follows when connecting the Gas In-Line-Filter (see Figure 3).
1. Turn off gas supply.
2. Cut gas hose between gas supply and incubator at desired point of Gas In-Line-Filter
integration.
3. Put one compression fitting on each gas hose’s end. The thread side of both compression
fittings faces towards the newly created end.
4. Connect each end of the gas hose to one coupling body and fix with one compression fitting.
5. Connect coupling bodies and Gas In-Line-Filter. A tight connection is confirmed by a “click”.
Pay attention to the direction of the gas flow. An arrow on the Gas In-Line-Filter shows the
direction of gas flow.
6. Open the shut-off valve and check for any gas leakages.
7. Check the gas pressure occasionally.

WARNING
Please check gas tightness of couplings.
In autoclave:
fractionated vacuum, 20 minutes at 121 °C or
fractionated vacuum, 3 minutes at 134 °C
We recommend this method for filling the humidification stone with suitable water. Place the humidification
stone into the provided gap of the Labo C-Top. Draw 8 ml - 9 ml of suitable water into a syringe and pour it
evenly on the humidification stone (see Figure 4). Overflowed water can be removed with a soft cloth.

The alarm statuses signalled on the device acoustically and visually may be forwarded for external
evaluation by means of the potential-free contact at the remote alarm output. The plug (REF 14787) must
be connected as shown in the picture and then connected to the remote alarm socket.
The switching statuses of the potential-free remote alarm are shown in the following pictures:

Interface RS485
The interface together with the optional software LaboDat+ is used for data capturing and evaluation on a
PC. Please contact us if you have questions concerning this software. We gladly provide further information
and will send you a non-binding offer.
The mains connection cable connects the device to the power supply. The device operates with
110 V - 230 V AC, 50/60 Hz.
WARNING: Care must be taken that the voltage given on the model
plate (on the back of the device) coincides with the supply voltage.
After the incubator is acclimatised to room temperature, it can be switched on with the mains switch.
Afterwards it takes about 30 minutes to reach set temperature of 37 °C inside. The default settings of the
device are 37 °C and a gas flow of 20 ml/min.
The gas supply is activated only when the set temperature is reached.

Set/Change Set-points
Hold down the menu key for two seconds to access the selection menu SETPOINT/OPTION. The individual
set-points can be accessed by repeatedly pressing >>>. Set-points are defined by means of the arrow
keys. Holding down the keys will change the respective set-point faster. When the setting is completed, the
new set-point is accepted by pressing >>> and permanently stored. The set-points remain in the memory
after the device is disconnected from power supply.
Set-point Temperature
The set-point for temperature should be selected between 30 °C and 42 °C. The selected set-point must
be at least 7 °C above the expected ambient temperature.
43
Labo C-Top Labotect
Set-point Flow
The set-point can be selected in the range of 20 ml/min to 30 ml/min in 5 ml/min increments. At a set-point
of 0 ml/min the gas flow is interrupted. After switching on the device, flow regulation only starts when the
set temperature is reached.
After every opening of the lid (lid is closed again), the incubation chamber is flushed with an increased gas
flow of 250 ml/min for 3 minutes.
Set-point Over-Temperature
The set-point can be selected between 30 °C to 45 °C. It should be selected at least 0.5 °C above the set
temperature value.
If the temperature inside the incubator exceeds the set-point for over-temperature due to a defect or due to
a high ambient temperature, the ALARM label blinks, a blinking A is shown on the right side of the display
and an alarm sound is switched on (see Figure 8). The alarm sound for over-temperature cannot be
switched off in the menu OPTION. To prevent further temperature increase, all heaters are switched off.
If the set-point for temperature is changed, the set-point for over-temperature must also be changed.
Optional Settings
In the OPTION menu, it is possible to switch off the alarm sound, select language settings, enter the device
number for capturing measuring values and define the tray number for unambiguous sample administration.
6.3.7.2 rH-alarm
The rH-alarm can be deactivated for each chamber separately. So, one chamber can be used without
humidification without giving rH-alarm. After each switching-on of the incubator, the rH-alarm is activated
for both chambers.
6.3.7.3 Language
The output on the display is made in a language selected by the user. It is possible to choose between
German (D) and English (GB).
44
Labotect Labo C-Top
If the set-point for gas flow cannot be reached even after a longer
period, the valve is blocked for reasons of safety. The valve is
unblocked again when the lid is opened.
